环境因子对龙牙百合脱毒组培苗移栽成活率的影响

摘    要:以万载县龙牙百合鳞茎诱导的组培苗为试材,采用正交实验分析了温度、湿度、光照等环境因子对移栽成活率的影响,以期明确环境因子对龙牙百合脱毒组培苗移栽成活率的影响。结果表明:1)单一环境因子中,龙牙百合移栽成活率随试验温度、湿度的升高均呈先升后降趋势,温度高于25℃时,移栽成活率达87.0%以上,湿度在70%~80%时有利于移栽的成活;随着试验光照强度的增强移栽成活率呈升高趋势,组培苗光照系统虽然较弱,但强光照能够促进光合系统发育,促进移栽成活率的提高;2)环境因子交互影响移栽成活,温度是龙牙百合组培苗移栽成活的主要因子,其次依次为湿度、光照强度,试验环境因子最优组合为温度25℃+湿度85%+光照强度30%、温度25℃+湿度75%+光照强度30%;3)全年中移栽成活率为20.0%~95.0%,成活率峰值出现在3、10月,7、8月不适宜移栽,龙牙百合组培苗应在上半年尽早移栽。因此,龙牙百合组培苗移栽时应在适宜生长的上半年尽早栽种,温度在25℃以上、基质湿度70%以上、不遮阴处理能够有效提高移栽成活率。

Abstract:The tissue culture seedlings of Lilium brownii var.viridulum induced by bulbs of Wanzai county were used as test material.The environmental factors such as temperature,humidity,and light on the survival rate were analyzed,in order to clarify the influence of environmental factors on the survival rate of virus-free tissue culture seedlings of Lilium brownii var.viridulum.The results showed that,1)in a single environmental factor,the survival rate of Lilium brownii var.viridulum increased first and then decreased with the increase of test temperature and humidity.When the temperature was higher than 25℃,the survival rate reached more than 87.0%.The humidity was 70%to 80%,which was beneficial to the survival of tissue culture seedlings.With the increase of the experimental light intensity,the survival rate was increased.Although the light system of tissue culture seedlings was weak,strong light could the development of photosynthetic systems and the survival rate.2)Environmental factors interactively affect the survival rate.Temperature was the main factor for the survival rate of Lilium brownii var.viridulum,followed by humidity and light intensity in order.The optimal combination of experimental environmental factors was‘temperature 25℃+humidity 85%+light intensity 30%’‘temperature 25℃+humidity 75%+light intensity 30%’.3)The survival rate in the whole year was 20.0%to 95.0%,and the peak survival rate appears in March and October.It was not suitable for transplanting in July and August.Tissue culture seedlings of Lilium brownii var.viridulum should be transplanted as early as possible in the first half of the year.Therefore,Lilium brownii var.viridulum tissue culture seedlings should be transplanted as early as possible in the first half of the year.The temperature was above 25℃,substrate humidity was above 70%,and non-shading treatment can effectively improve the survival rate.
