桑原生质体分离技术的研究

摘    要:用桑子叶、桑幼叶、悬浮培养细胞、愈伤组织四种材料,分别进行了原生质体的分离方法及有关条件的研究。结果在适宜酶溶液浓度下,桑原生质体的释放速度和产量顺序为,桑子叶>幼叶>悬浮培养细胞>愈伤组织。酶液最适渗透剂浓度为0.5-0.6M的甘露醇。经预培养处理的材料,原生质体产量比对照提高12.9倍。

Abstract:Mulberry cotyledons,young leaves,suspension cells and calli were used for proto-plast isolation studies. The results showed that :1) Under appropriate enzyme concentra-tion,the release speed and yield of mulberry protoplasts was in the order of cotyledons>young leaves > suspension cells>calli;2) The optimum osmotic concentration forenzyme solution was 0.5~0.6M tnannitol;3) Pretreatment to young leaves increasedprotoplast yield by 12. 9 times.
