摘    要:参照国内外方法,选择M基因的保守序列作为靶序列,应用快速、高效、特异的一步法RT-PCR检测法进行禽流感病毒型特异性诊断,得到大小为229bp的特异性产物,与预期结果一致.另外,采用柱式总RNA提取方法,与酚/氯仿的抽提方法相比,具有快速,对操作人员技术要求较低,整个反应和扩增体系设计合理等特点.以上技术,简化了操作程序,节约了检测时间,缩小了因微量操作引起的误差,可获得较理想的结果和可靠的检测数据.

Abstract:According to the PCR technology based on the latest research development at home and abroad,choosing the M gene conservative sequence as a target sequence,the fast,efficient,specific one-step RT-PCR method was used for detecting avian influenza virus.A 229 bp specific product was obtained,which is consistent with the anticipated result.In addition,the column method was used for total RNA extraction.Compared with the phenol/chloroform extraction method,the column method was fast and simple,and the entire amp...
