摘    要:应用银染法AFLP鉴定以父本‘多萼朱砂”与母本‘雪梅’杂交所得 12株F1 子代。以E AT +P AT选择性引物对扩增 ,父本 180bp的特异带在 12株F1 代中都呈阳性带 ;母本 170bp和 171bp两条特异带在 12株F1 代中也都呈阳性带 ,双亲基因组DNA中这 3个特异位点在F1 子代中稳定遗传。结果表明 ,这 12株均为‘多萼朱砂’ב雪梅’的F1 子代 ,AFLP是适合梅花亲子鉴定的简便技术。AFLP对梅花杂交选育新品种的早期选择具有应用前景。

Rapid Identification of Parentage in Prunus mume by AFLP Assay
Abstract:Parentage determination of 12 individuals from the cross Prunus mume `Xue Mei'×Prunus mume `Duo'e Zhusha' was conducted using the silver staining AFLP method. The results of amplification in F 1 population with the selective primer combination of E-AT + P-AT were shown as follows: specific band of 180 bp of the male parent was present in all the 12 progenies of F 1 population and two specific bands of 170 bp and 171 bp of the female parent were present in all the 12 progenies too. And these three bands could inherit from parents to progenies of F 1 population constantly. It is concluded that these 12 individuals are all hybrids. Therefore, AFLP is a rapid and simple method for parentage determination in Prunus mume. It will be useful for early selection of hybrids in cross breeding of Prunus mume.
