Abstract:In order to identify the etiology of outbreak deaths of mandarinfish,a dominant bacterial strain(ZQ202010)was isolated from the infected mandarinfish.This study identified the pathogens through clinical necropsy observation,bacterial isolation and culture,Gram staining observation,biochemical identification,16S rRNA and gyr B gene sequencing analysis,animal regression test,virulence gene detection and drug sensitivity test.The results showed that the strain ZQ202010 was a Gram-negative microbacillus.Biochemical identification results showed that the strain was motile,and lysine decarboxylase,oxidase and VP indole were positive.The reaction of arginine hydrolase and hydrogen sulfide was negative.The 16S rRNA and gyr B gene sequences analysis showed that the isolate and Aeromonas veronii covered into one branch,and the homology was more than 99%.The result of animal regression showed that the LD50 of the isolate to mandarinfish was 4.09×106 CFU,showing high virulence to mandarinfish.The amplification result of virulence gene showed that the isolate carried 7 virulence genes including aer A,Alt,Fla,Exu,Ser,ahy B and Lip,which further indicated that the isolate had high virulence.Drug susceptibility test showed that cefradine,temicocacin,ceftriaxone,florfenicol,doxycycline and other drugs had the most inhibitive effects on the strain.At the same time,it showed resistance to ciprofloxacin,cefotaxime,penicillin,amoxicillin/clavulanic acid,enrofloxacin,cotrimoxazole,rifampicin and ofloxacin.Therefore,a pathogenic Aeromonas veronii strain was isolated in this study,which provides a scientific basis for the prevention and control of Aeromonas vinieri from Siniperca chuatsi.
