鸡传染性法氏囊病病毒的分离鉴定与培养特性研究

摘    要:为研究鸡传染性法氏囊病病毒在不同宿主上的培养特性,从河南省某疑似感染鸡传染性法氏囊病病毒的鸡场采集病料,通过琼脂免疫扩散试验初步筛选出IBDV毒株,并将毒株回归易感鸡,在SPF鸡胚及DF1细胞上培养。结果显示:培养出的病毒作为抗原与标准阳性血清之间均出现阳性沉淀线;回归SPF鸡试验显示发病率达100%,死亡达60%,病死鸡只出现法氏囊肿大、出血甚至出现紫葡萄样;该毒株能引起鸡胚CAM增厚、胚体出血,引起DF1细胞变圆脱落;ELD50与TCID50分别为10-4.6/0.1 mL、10-5.5/0.1 mL。试验证实IBDV分离株,属于超强毒株,该毒株可以在鸡胚和DF1细胞上适应增殖,将其命名为HN12号分离株。

Abstract:To study the cultivate characteristics in different host, An IBDV strain was selected preliminary with Agar diffusion test, which collected from a chicken farm suspected infection with infectious bursal disease virus in Henan Province. The strain were infected susceptibility chicken, SPF chicken and DFI cells. The results showed that, there were positive precipitation line in AGID; This strain could cause SPF chickens (morbidity with 100% and mortality with 60%) bursal swelling, bleeding, and even purple grape-like were showded; cause chick embryo CAM thickening, chicken embryos bleeding, cause the DF1 cells round off, the ELDso and TCID~o was respectively 10-4.6/0.1 mL and 10-5.5/0.1 mL. The results confirmed that the isolated IBDV stain belongs to vvlBDV, it could replicate in chicken embryo and cell. It was named HN12.
