摘    要:以携带广谱持久抗稻瘟病基因Pi9的籼稻品系75-1-127为供体亲本,以优质常规晚稻湘晚籼11号为受体亲本及轮回亲本,利用Pi9基因内特异标记,在连续回交自交群体中开展分子标记辅助选择,定向改良湘晚籼11号的稻瘟病抗性。24份稻瘟菌菌株室内接种及田间病圃抗性鉴定结果表明,75-1-127和湘晚籼11号的抗菌谱差异显著,抗性频率分别为91.7%和29.2%,75-1-127高抗苗瘟和穗瘟,而湘晚籼11号高感苗瘟和穗瘟;鉴定出1个稳定高效的显性DNA标记Ins2-1,该标记引物从75-1-127基因组中扩增出1条532 bp的清晰条带,而在湘晚籼11号基因组中无扩增产物。利用Ins2-1开展标记辅助选择育种实践,从湘晚籼11号×75-1-127的BC6F3群体中鉴定出9个抗病改良系,为进一步培育优异抗病水稻新品种打下了基础。

Abstract:Pi9 is a cloned broad-spectrum and durable blast resistance gene.To improve blast resistance of the indica rice variety Xiangwanxian 11,Pi9-specific DNA marker was employed for molecular marker-assisted selection breeding in the consecutive backcrossing and self-pollination populations of Xiangwanxian 11×75-1-127. Resistance responses to 24 tested M.oryzae strains were quite different between the Pi9 donor 75-1-127 and the receptor Xiangwanxian 11,presenting 91.7% and 29.2% resistance ratio respectively in the greenhouse inoculation assay,and 75-1-127 showed high-level seedling and panicle blast resistance but Xiangwanxian 11 was highly susceptible in field nursery. An efficient PCR-based dominant marker Ins2-1 was identified,which can amplify a stable 532 bp band from 75-1-127 genome but nothing from that of Xiangwanxian 11. Nine improved blast resistant rice lines were confirmed from the BC6F3 population of Xiangwanxian 11×75-1-127 using the marker Ins2-1,which paved a way for further breeding elite rice varieties.
