| 摘 要： | 目的]确定穿龙薯蓣花粉活力及柱头可授性的快速、有效测定方法。方法]分别采用I2-KI染色法、TTC染色法、亚甲基蓝染色法、MTT染色法和无机盐培养法测定穿龙薯蓣的花粉活力,采用联苯胺-过氧化氢法和MTT染色法测定其柱头的可授性。结果]I2-KI和TTC染色法对穿龙薯蓣花粉的染色效果不理想,不适于观测其花粉活力;亚甲基蓝染色法使花粉生活力的测定值偏高;MTT染色法使花粉细胞出现紫色、淡红色、无色3种颜色反应,利于分辨花粉生活力的有无。联苯胺-过氧化氢法不适于观测穿龙薯蓣柱头的可授性;MTT染色法能较准确地观测出不同时期柱头可授性的动态变化。结论]MTT染色法适于检测穿龙薯蓣的花粉活力及柱头可授性。
