干旱和光联合胁迫对长春花CesA基因和Pme基因表达的影响

摘    要:以长春花叶片为材料,通过同源序列扩增法,获得与细胞壁合成相关的纤维素合成酶(Cellulose synthase,CesA)基因和与细胞壁重要组成部分果胶的变化密切相关的果胶酯酶(Pectin methylesterase,Pme)基因,利用定量的RT-PCR,对干旱和光联合胁迫下CesA基因和Pme基因的表达情况进行了研究。结果表明:干旱胁迫条件下,CesA基因表达稍有减弱,Pme基因表达稍有增强。光胁迫条件下,CesA基因表达明显弱于正常光条件,呈逐渐减弱趋势,CesA基因表达与光胁迫的关系为负调控;而Pme基因表达则明显强于正常光条件,呈现增强趋势,Pme基因表达与光胁迫的关系为正调控。说明光胁迫是CesA基因和Pme基因表达变化的主要影响因素。在干旱和光联合胁迫下,CesA基因表达整体减弱,Pme基因表达整体较强。

Abstract:Cellulose synthase (CesA) gene and Pectin methylesterase (Pme) gene, which are related with the exoskeleton of cell wall and the degradation of cell wall by hydrolyzing pectin were obtained by sequence amplification method using the leaves of Catharanthus roseus (L.) G. Don as experimental materials. The expressions of CesA and Pme genes in leaves of C. roseus were investigated under combined stresses of drought and light. Results revealed that the expression of CesA gene from C.!roseus was slightly weak and the expression of Pme gene was slightly stsong under drought stress. The expression of CesA gene under light stress was significantly weaker than that under common light condition, showing a gradually decreasing tendency. A good negative correlation was observed between CesA gene expression and light stress. The expression of Pme gene under light stress was significantly stronger than that under common light condition, showing a gradually increasing tendency. There was a positive correlation between Pme gene expression and light stress. It indicates that light stress is the main factors influencing the expressions of CesA and Pme genes. The expression of CesA gene weakened and that of Pme gene enhanced under the combined stresses of drought and light.
