IGF1R和IGF2R基因在鸭脂肪组织中的发育表达模式比较

摘    要:为比较胰岛素样生长因子1受体(IGF1R)和胰岛素样生长因子2受体(IGF2R)在鸭脂肪组织发育中的作用,利用RT-PCR技术,扩增出鸭IGF1R、IGF2R部分编码区序列,并采用荧光定量PCR检测IGF1R、IGF2R基因mRNA丰度在1~8周龄鸭腹脂、背皮和肝脏中的表达变化。成功获得了鸭IGF1R和IGF2R部分序列,长度分别为798 bp和716bp。腹脂中IGF1R和IGF2R基因的表达都呈先上升后下降的变化规律,4周龄时表达量最高;皮下脂肪中IGF1RmRNA的表达规律是在1周龄时极显著高于其他各时间点(P<0.01),2周龄时显著下降,随后缓慢上升到4周龄时达到最高,之后又逐渐下降。IGF2R mRNA的表达水平呈先上升后波动下降的趋势,以4周龄时表达量为最高,5~8周龄时波动下降;肝脏中IGF1R和IGF2R基因的表达规律相似,表达量总体都呈现1周龄后急剧下降而后平稳的趋势。综合上述结果可知,IGF1R和IGF2R基因对肝脏的脂肪合成以及肝外脂肪组织增殖、分化的调控具有差异性。

Abstract:The insulin-like growth factor 1 receptor and insulin-like growth factor 2 receptor were studied to investigate the different genetic mechanism of these two genes in duck adipose tissue development.The partial coding domain sequence of duck IGF1R and IGF2R were amplified by RT-PCR and then the developmental expression patterns of IGF1R,IGF2R genes in abdominal fat,subcutaneous fat and liver in duck during the stage from week 1 to week 8 were analyzed using real-time quantitative PCR.Results showed that the length of the IGF1R and IGF2R sequence were 798 bp and 714 bp,respectively.That developmental expression patterns of IGF1R and IGF2R in abdominal fat were similar,which were increased first and then declined,it were relatively higher at 4 weeks.In subcutaneous fat,the expression level of IGF1R mRNA was significantly higher at 1 week than it at other weeks,rapidly declined at 2 week,obviously increased at 4 week,and then decreased slowly.The expression level of IGF2R mRNA exhibited down-regulation,but a little fluctuant after up-regulated,it was relatively higher at 4 week,then declined with a little fluctuant at 5 to 8 weeks.The developmental expression patterns of IGF1R and IGF2R in liver were similar to show quickly decreased after 1 week,and then maintained steady.The results suggest the regulation mechanism of these two genes in the lipogenesis of liver and the proliferation and differentiation of fat cells were different.
