摘    要:通过不同处理和多种影响因素对不同类型小麦花药培养效应的比较研究表明,旱作春小麦花药培养愈伤组织诱导率和绿苗分化率均存在显著的基因型差异,其中以普通小红麦诱导率最高,达8.12 %,八倍体小黑麦中以20-1528诱导率最高,达7.71 %。诱导培养基中2, 4-D、水解乳蛋白和蔗糖之间的适宜配比为:2.0∶0.5∶90(g/L);而在分化培养基中的适宜配比为:0∶0.5∶70(g/L)。对花药进行2 d低温预处理和32 ℃高温培养具有较高的花药愈伤诱导率,分别达7.6 % 和6.58 %。

Abstract:Effects of different treatments and a few extrafactors on the high efficiency anther culture of different genotypes of spring wheat in dryland were studied. The results showed that both induction percentage of pollen callua and calli giving green plantlets in the high efficiency anther culture of spring wheat in dryland varied with the genotypes,among which the induction percentage of pollen callus of the common small red wheat had reached 8.12 % as the highest level in comparison with others varieties of common wheat; while the induction percentage of callus of the strain 20-1528 had reached 7.71 % as the highest among the strains of octploid rye. The fittest proportion of 2, 4-D, LH and sucrose in the best-induced medium was 2.0∶0.5∶90 (g/L),but in the best medium of differentiation, the fittest proportion was 0∶0.5∶70 (g/L). Meanwhile, higher induction percentage of pollen callua had been obtained with low temperature pretreatment for two days and high- temperatured culture at 32℃, and reached 7.6 % , 6.58 % separately.
