甘蓝亲本防退化技术研究

摘    要:目的]探索甘蓝亲本防退化技术。方法]以“春魁”甘蓝父母本为材料,选取培养10d后的无菌苗茎尖为诱导材料镑种在培养基上进行诱导培养、愈伤组织继代培养、芽分化培养、生根培养和炼苗移栽,研究采用茎尖组培快繁技术繁殖甘蓝原种。结果]诱导出的不定芽在2种分化培养基内幼苗长势均良好,在MS+6-BA2.0mg/L+NAA0.2mg/L培养基中幼苗增殖倍数为3—5倍,且生成大量根系:在MS+6-BA2.0mg/L培养基中幼苗增殖倍数可达到5~7倍,没有根系生成。2个亲本利用组培繁殖植株的种子产量高于常规繁殖植株的产量,9701—2和HT502亲本组培繁育的种子产量分别比常规繁殖的高16.8%和39.1%。结论]采用茎尖组培快繁技术繁殖甘蓝原种,可减少甘蓝原原种用量,提高繁殖系数和种子发芽率。
