摘    要:目的]优化蕨菜Pteridium aquilinum (Linn.) Kuhn var.latiusculum(Desv.)Underw.exHeller]总黄酮的提取工艺,为蕨菜黄酮类化合物的开发利用提供理论依据。方法]在单因素试验基础上,以总黄酮得率为考察指标,采用正交试验对影响蕨菜总黄酮提取的因素,乙醇体积分数、沸腾时间、淋洗时间和料液比进行优化。结果]蕨菜总黄酮最佳提取工艺为:乙醇体积分数60%,沸腾时间2.0 h,淋洗时间1.0 h,料液比1∶30(W/V)。在此优化工艺条件下,蕨菜总黄酮得率达8.80%。蕨菜不同器官中总黄酮含量趋势为:脱脂叶片〉叶片〉脱脂叶柄〉叶柄。结论]该研究建立的提取蕨菜总黄酮的方法得率高于传统方法,且节省时间、简单易行。

Optimization of Extraction Process of Flavonoids from P. aquilinum (Linn.) Kuhn var. latiusculum (Desv.)Underw.ex Heller

Abstract:Objective]The research aimed to extraction process of flavonoids from P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller,to provide the theoretical basis for development and utilization of P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller flavonoids.Method]Based on the single factor test,taking the extraction rate of flavonoids as index,the orthogonal test was used to optimize the factors influencing flavonoids extraction from P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller,such as volume fraction of ethanol,boiling time,leaching time,and the ratio of solid to liquid.Result]The results showed that the best extraction process of flavonoids from P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller was as follow:volume fraction of ethanol 60%,boiling time 2.0 h,leaching time 1.0 h,and the ratio of solid to liquid 1:30.The extraction rate of flavonoids was 8.80% under the optimized extraction process.The flavonoids contents in different organs of P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller was in order: Fat-free leaf> leaf> fat-free petioles> petioles.Conclusion]The establishing method of extracting flavonoids from P.aquilinum(Linn.) Kuhn var.latiusculum(Desv.)Underw.ex Heller in this study has higher extraction rate comparing with traditional methods,and it is characteristics of saving time,simple and easy to use in practice.
