摘 要: | 目的]探究水稻纹枯病拮抗细菌的抑菌活性成分的发酵制备条件。方法]通过点接种于平板与水稻纹枯病对峙生长对水稻纹枯病拮抗细菌进行了初筛,通过摇瓶发酵进行了复筛,并通过形态鉴定及16S RNA测序构建系统发育树对所得菌株进行鉴定;通过单因素试验研究了所得菌株的抑菌活性成分的发酵制备条件。结果]初筛获得17株水稻纹枯病的拮抗细菌,复筛获得菌株Lys-67,该菌株的发酵液对水稻纹枯病生长具有明显的抑制作用,菌株Lys-67鉴定为枯草芽孢杆菌(Bacillus subtilis)。经单因素试验,菌株Lys-67利用麸皮5.0%,于35℃、摇床转速200 r/min条件下发酵3 d,经牛津杯打孔法检测,150μl发酵液中活性成分抑制水稻纹枯病的抑菌圈达36 mm。结论]为菌株Lys-67的进一步研究及生防应用奠定了基础。

Abstract: | Objective] The aim was to study the fermentation conditions of antagonistic bacteria active ingredient against Rhizoctonia solani.Method] Some antagonistic bacteria against R.solani were screened through point inoculating contrary to R.solani on agar petri plates and shaking flask fermentation; the obtained strain was identified through morphological identification and 16S RNA sequencing to construct phylogenetic tree ; and fermentation conditions of active ingredient of the obtained strain against R.solani were studied through single factor experiment.Result] Seventeen strains of bacteria antagonist R.solani were obtained,and strain Lys-67 was obtained finally; strain Lys-67 was identified as Bacillus subtilis.Also,150 μl fermentation broth of strain Lys-67 resulted in 36 mm of inhibition zone against R.solani,determining by method of drilling hole with Oxford cup on agar plate,while strain Lys-67 was cultured in wheat bran 5.0%,at 35 C,rotation speed 200 r/min for three days.Conclusion] The results lay basis for further study and application of strain Lys-67. |
