摘    要:采用响应面法对木聚糖酶产生菌变种米曲霉(Aspergillus oryzae)RM-503的发酵培养基进行了优化。用Plackett-Burman法(Plackett-Burman Design,P-B)对影响变种米曲霉产木聚糖酶发酵的相关因素进行了评价。筛选有显著效应的2个因素:KH2PO4和ZnSO4.7H2O。在此基础上,采用最陡爬坡路径逼近最大响应区域,利用中心旋转组合设计及响应面分析确定影响木聚糖酶产量的关键因素KH2PO4和ZnSO4.7H2O的最佳水平。经过上述优化,木聚糖酶产量比基本培养条件下提高了1.399倍,达到8.681 U/mL。

Abstract:The fermentation medium of Aspergillus oryzae RM-503 producing xylanase is optimized by using response surface methodology(RSM).In the first step of optimization,with Plackett-Burman design,KH 2 PO 4 and ZnSO 4 ·7H 2 O are found to be the important factors affecting xylanase production significantly.Then the steepest ascent experiment is adopted to approach the optimal region of the medium composition.At last,the central composition design and response surface methodology are used to determine the optimal levels of the main factors.After optimization,the yield of xylanase production is increased by 139.9% compared with that in the original medium.
