| 摘 要： | 目的]采用5种方法对不同品系的油菜花粉生活力进行测定,以找出油菜花粉生活力的最佳测定方法。方法]5种花粉生活力测定方法分别为蔗糖萌发测定法、醋酸洋红染色法、氯化三苯基四氮唑(TTC)染色法、碘—碘化钾染色法和红墨水染色法。结果]TTC染色法和红墨水染色法测定值偏高,碘-碘化钾染色法测定结果偏低;蔗糖萌发测定法和醋酸洋红染色法测定结果比较接近,是测定油菜花粉生活力的最佳方法。结论]可将蔗糖萌发测定法和醋酸洋红染色法结合起来,先通过醋酸洋红染色法掌握花粉的基本存活情况,之后进一步用蔗糖萌发测定法测定其花粉的萌发能力,这有利于正确地反映花粉生活力。

| Abstract: | Objective]Five methods were used to determine the pollen vitality of different lines rape to find out their best determination method.Method]Five determination methods were sucrose-based germination method,acetocarmine dyeing method,TTC dyeing method,I2-KI dyeing method and red ink dyeing method,respectively.Result]The results showed that the pollen vitality values of different lines rape determined by red ink dyeing method and TTC dyeing method were on the high side,while that was on the low side determined by I2-KI dyeing method.The determination rescult of sucrose-based germination method and acetocarmine dyeing method were closer,and were the best way to determine the pollen viability.Conclusion]When determining the pollen vitality of rape,it is better to first grasp the basic survival situation of pollen by acetocarmine dyeing method,and then determine its germination vigor by sucrose-based germination method,which is in favor of correctly reflecting pollen vitality. |
