摘    要:目的:建立商洛桔梗药材的高效液相色谱(HPLC)指纹图谱,为商洛桔梗药材的道地性研究提供可靠资料。方法:采用HPLC法测定来源于商洛7个县(区)的39个桔梗样品。色谱条件:Hypersil C18柱(250 mm×4.6 mm,5μm),以乙腈和H3PO4混合溶液作为流动相,采用梯度洗脱方式,流速为0.5 mL/min,检测波长为210 nm,柱温30℃。结果:建立了39批桔梗药材的HPLC指纹图谱,找到18个共有特征峰。结论:该方法简单,重现性好,可以用于商洛桔梗药材的品质鉴定与评价。

Abstract:This study aimed to build the high performance liquid chromatography (HPLC) fingerprint of authentic medicinal material Platycodon grandiflorum in Shangluo, and to provide reliable data for the geoherbalism study of this medicinal material. Thirty - nine Platycodon grandiflorum samples from 7 counties (districts) of Shangluo city were determined by using HPLC under the following chromatographic conditions: using Hypersil C18 column (250 mm × 4.6 mm, 5μm), mobile phase being the mixed solution of aeetonitrile and H3 POa, adopting gradient elution mode, flow velocity being 0.5 mL/min, detecting wavelength being 210 nm,and column temperature being 30℃. Finally, we built the HPLC fingerprint of 39 batches of Platycodon grandiflorum, and found out 18 common characteristic peaks. It was concluded that this method was simple with good repeatability, and could be used for the identification and evaluation of authentic medicinal material Platycodon grandiflorum in Shangluo.
