六出花瓶插过程中乙烯代谢特征及其调控效应

摘    要:为了探究六出花瓶插过程中乙烯代谢特征及其调控效应,为六出花切花保鲜剂的研发提供理论依据,以花苞期的六出花切花为供试材料,在基础瓶插液( 1 %蔗糖 + 200 mg·L-1 8-羟基喹啉)中分别加入20 mg·L-1乙烯利(ETH)、4 mg·L-1纳米银(NS)、75 mg·L-1水杨酸(SA)和10 mg·L-1精胺(SP)配制瓶插液处理六出花切花,分析瓶插过程中切花形态、生理和基因表达3个层面的指标变化。结果表明:(1)含有乙烯利的瓶插液处理显著缩短了六出花切花瓶插寿命,含有纳米银、水杨酸、精胺的瓶插液处理均能延长六出花切花瓶插寿命,其中以含有4 mg·L-1纳米银的瓶插液处理效果最佳,延长了瓶插期 2 d。(2)含有乙烯利的瓶插液处理显著加速了花枝鲜样质量和花瓣可溶性糖、可溶性蛋白含量的降低,增加了花瓣丙二醛含量和相对电导率,显著提高了花瓣乙烯代谢水平。含有纳米银、水杨酸、精胺的瓶插液处理六出花切花均不同程度地延缓了花枝鲜样质量和花瓣可溶性糖、可溶性蛋白含量的降低,抑制了丙二醛含量和相对电导率的升高,降低了乙烯代谢水平。(3)含有乙烯利的瓶插液处理增加了开花各时期乙烯合成及信号转导关键基因(AlACO、AlACS、AlCTR1、AlERS1AlEIN3)的表达量;纳米银、水杨酸和精胺均能不同程度地抑制乙烯合成及信号转导关键基因的表达。研究结果显示六出花为乙烯末期上升型花卉,外源施加乙烯抑制剂可以延缓六出花切花衰老,提高瓶插品质,其中以4 mg·L-1的纳米银效果最佳。

Abstract:In this paper, the characteristics of ethylene metabolism and regulatory effect of Alstroemeria hybrida cut flowers during vasing life was studied to provide a theoretical basis for the research and development of A. hybrida cut flowers preservative. Cut flowers of A. hybrida in bud period were treated by adding 20 mg·L-1 ethephon (ETH), 4 mg·L-1 nano-silver (NS), 75 mg·L-1 salicylic acid (SA) and 10 mg·L-1 spermine (SP), respectively into basic vase solution (1% sucrose + 200 mg·L-1 8-hydroxyquinoline), and the morphological, physiological and gene expression changes of cut flowers during vase period were analyzed. The results were as follows: (1) Ethephon vase treatment significantly shortened the vase life of A. hybrida flowers. Treatments with nano-silver, salicylic acid and spermidine could extend the vase life of A. hybrida cut flowers. The treatment with 4 mg·L-1 nano-silver had the best effect, and the vase life extended for two days. (2) The treatment with ethephon significantly accelerated the decrease of fresh quality of flower branches, soluble sugar and soluble protein contents of petals, increased MDA content and relative electrical conductivity of petals, and significantly improved ethylene metabolism of petals. The vase treatment containing silver, salicylic acid and spermine delayed the decrease of fresh quality of flower branches, soluble sugar and soluble protein contents of flower petals, inhibited the increase of MDA content and relative conductivity, and significantly reduced ethylene metabolism. (3) Vase treatment with ethephon increased the expression levels of keygenes for ethylene synthesis and signal transduction (AlACO, AlACS, AlCTR1, AlERS1 and AlEIN3) in each
