农杆菌携带柞蚕抗菌肽基因转入桑树的研究

摘    要:人工合成柞蚕抗菌肽D基因通过根癌农杆菌Ti质粒载体转入桑树叶盘,获得基因转化工程苗。柞蚕抗菌肽对桑树青枯病假单孢菌有明显杀菌作用。人工合成柞蚕抗菌肽基因导入农杆菌感染桑树叶盘并诱导成苗。经卡那霉素筛选,胭脂碱电泳检测及抗菌肽基因探针进行印迹点杂交,确认抗菌肽基因转化桑苗成功,为培育抗青枯病品种打下基础。

Abstract:Transfering of mulberry plant ,Morus alba by infection of Ti plasmid from Agrobac-terium tumefaciens was studied. Plants selected on kanamycin containing medium wereconfirmed to express the introduced 35S-Synthetic antibacterial peptide gene(ABG)fromChinese oak silkworm, Antheraea pernyi.The introduced gene was obtained by fusionconsisted of CaMV 35S promoter with ABG. Using the kanamycin resistant gene in Co24plasmid. The mulberry leaf discs of advantition bud formed from infect tissues were se-lected on the MS medium containing kanamycin at 25μg/ml.When they were transferedto the selection medium at 4一5days,the nopalin synthetic enzyme activity was detectedby electrophoresis. The transformation was confirmed in 40 days with kanamycin resis-tance by DNA dot blotting. The results showed the synthetic antibacterial peptide genewas transfered into mulberry plant.
