滤光膜对黄檗茎段愈伤组织诱导及3种生物碱含量的影响

摘    要:以黄檗茎段为外植体,研究不同颜色滤光膜对其愈伤组织诱导及生物碱(小檗碱、掌叶防己碱、药根碱)含量的影响。将接种到MS+1.5 mg/L BA+0.5 mg/L NAA+20 g/L葡萄糖培养基上的茎段置于不同条件下培养,20天后对其进行形态观察并取样用于生物量测量及生物碱含量测定。结果表明,各种处理条件下外植体均100%的诱导了愈伤组织,但不同处理对外植体状态、愈伤组织生长和次生代谢产物含量均有不同程度影响。其中对照、蓝膜处理下诱导的愈伤组织较疏松、颜色鲜艳、生长旺盛,而红膜、绿膜和黄膜则诱导出较少愈伤组织且呈老化状态。进一步研究表明,对照和蓝膜处理下3种生物碱含量明显高于其他处理。实验表明,对照和蓝膜对愈伤组织生长和次生代谢产物含量的积累效果最好,黄膜次之,绿膜和红膜最差。
