丽格海棠的离体培养及快速繁殖

摘    要:为了探索丽格海棠杂合体母株优良性状的保存方法,以便快速有效地进行丽格海棠种苗的生产,以丽格海棠的叶片做外植体进行了离体培养和快速繁殖研究。经实验筛选出各培养阶段最适宜的培养基:①芽分化培养基为MS+6-BA 2.0 mg/L+NAA 0.2 mg/L+GA30.2 mg/L;②芽增殖培养基为MS+6-BA 1.0 mg/L+NAA 0.5 mg/L;③生根培养基1/2MS+NAA 0.5 mg/L,上述培养基均添加2.5%蔗糖,0.8%琼脂,pH值为5.8。

Abstract:The foliage of rose crabapple(Begonia mannii L) was studied as explants for the preservation of the good characteristics of the haploid female line and the production of the seedlings of rose crabapple effectively.The optimum medium of bud differentiation was MS 6-BA 2.0 mg/L NAA 0.2 mg/L GA_3 0.2 mg/L.The optimum medium of bud propagation was MS 6-BA 1.0 mg/L NAA 0.5 mg/L.The optimum medium of inducing root was 1/2MS NAA 0.5 mg/L.Sucrose of 2.5% and agar powder of 0.8% were added in all mediums.The pH value was 5.8.
