摘    要:【目的】获得高抗水平的转拟南芥NPR1恢复系水稻253株系,比较转基因水稻与其非转基因野生型抗病性和农艺性状的差异,以期为转基因抗病育种提供材料。【方法】以通过连续自交获得的转AtNPR1基因253 T3代纯合株系和非转基因253为材料,于分蘖期采用剪叶法接种白叶枯病菌菌株13751于水稻叶片,14 d 后,对稻株发病情况进行调查;收获期考查6个转基因株系的农艺性状。【结果】6个转基因T3代株系植株的抗病性可稳定表达,其病斑长度均显著短于非转基因野生型253;253-3的抗病能力比野生型253增强50%以上,其他 5个株系253-4、253-7、165、166、167对水稻白叶枯病表现出高抗水平,抗病能力增强90%以上。与野生型253相比,转基因株系植株生长正常,不同株系间部分农艺性状无明显变化规律,而部分农艺性状显著升高或降低;株系166的株高、穗长、有效穗数、一次枝梗数、每穗总粒数、每穗实粒数和单株产量等显著优于非转基因恢复系253。【结论】拟南芥NPR1基因可作为培育广谱抗病作物的热门候选基因,株系166可作为水稻抗病育种的新种质。

Abstract:Objective]The present experiment was conducted to attain transgenic Arabidopsis thaliana NPR1 gene restorer rice line 253 with high resistance,and to compare the differences in agronomic traits among transgenic rice and wild type 253 to provide transgenic material for disease resistance breeding.Method]Six T3 generation lines(253-3,253-4,253-7,165,166 and 167) of transgenic rice cultivar 253,over-expressing A.thaliana AtNPR1 gene and wild type 253 were used for investigating their resistance against rice bacterial blight disease after inoculation for 14 days using leaf-cutting method at tillering stage and comparing their agronomic traits at harvesting stage.Result]Six T3 generation lines of transgenic rice cultivar 253 showed stable rice bacterial blight disease resistance,and their lesion lengths were significantly shorter than that of non-transgenic wild 253.As compared to wild 253,the disease resistance of line 253-3 enhanced over 50%,and that of the other five lines,viz.,253-4,253-7,165,166 and 167 enhanced over 90%.All of T3 generation lines showed normal growth and there were no significant regular changes in their agronomic traits,however,some agronomic traits showed significant increase or decrease compared to wild 253.The plant height,panicle length,effective panicles,primary rachis branches,total grains per panicle,full grains per panicle and yield per plant were significantly higher than that of non-transgenic 253.Conclusion]A.thaliana NPR1 gene is good candidate gene for breeding broad disease resistance crops in the future,and transgenic line 166 could be used as new material in breeding.
