斑马鱼CDKAL1基因的生物信息学分析

摘    要:目的]深入了解斑马鱼CDKAL1基因的结构特点。方法]运用生物信息学方法对斑马鱼CDKAL1基因的基因序列、编码蛋白的理化性质、结构及功能进行分析。结果]分析结果显示,此基因一部分存在于细胞质,并存在一个跨膜结构域,没有信号肽。该基因编码蛋白质的变异不大;含有3个功能结构域,均为Erk D-domain,分别位于L189L、384、V315;在Y292位置有一磷酸化位点;有UPF0004超家族、Radical SAM超家族和MiaB结构域3个保守结构域;在脑、肌肉、肾脏中表达。推测该基因表达的蛋白质可能是酶、异构酶或者中间代谢产物,与胰岛素受体的作用有关。结论]该研究为今后进一步研究人类Ⅱ型糖尿病提供了科学依据。

Abstract:Objective] The aim was to understand the structural characteristics of CDKAL1 gene from Danio rerio deeply.Method] The sequence of CDKAL1 gene from D.rerio and the physical and chemical properties,structure and function of its encoding protein were analyzed using the bioinformatics method.Result] The analysis result showed that the gene existed in the cytoplasm partially,had a trans-membrane domain and without signal peptide.The protein encoded by the gene had no significantly variation;it had 3 functional domains which was all Erk D-domain and located at L189,L384 and V315,resp.;there was a phosphorylation site at the location of Y292;the protein had 3 conserved domains of UPF0004 superfamily,Radical SAM superfamily and MiaB domain;it expressed in brain,muscle and kidney.It was conjectured that the protein expressed by the gene could be enzyme,isomerase or intermediate metabolite and it had relation with the function of insulin receptor.Conclusion] This research provided the scientific basis for the further study of human Type 2 diabetes for the future.
