摘    要:EDS1(Enhanced Disease Susceptibility 1)在植物抗病过程中起着关键的作用。为进一步研究其在调控茄子抗青枯病中的作用,根据茄子基因组序列设计引物克隆了EDS1,将其命名为SmEDS1。生物信息学分析表明SmEDS1最大开放阅读框包含1 809 bp,编码602个氨基酸残基。其包含该基因家族典型的LP结构域和EP结构域。为了进一步验证其功能,通过VIGS技术沉默茄子抗青枯病材料SmEDS1后接种青枯病病原菌,7 d后植株枯萎,结果表明EDS1正调控茄子抗青枯病。半定量结果表明,SmEDS1沉默后对其他信号基因的表达量有着显著影响,其中MAPK6、RAR1的表达量上调,而MAPK3、SIPK、PAD4、SGT1、TGA、EDR1、NPR1、ICS1、GLUA、EIL1和HSP90等基因的表达量下调,EBF2和ACO5的表达量不受SmEDS1的调控。此结果表明EDS1在调控茄子抗青枯病中起着重要的作用。

Abstract:EDS1 (Enhanced Disease Susceptibility 1)plays a  key  role  in  plant  disease  resistance.  To  further  analyze the EDS1 function in the eggplant bacterial wilt resistance, SmEDS1 was cloned according to the eggplant genome sequence. The bioinformation analysis showed that the max ORF box  of  SmEDS1  was  1809 bp  which  coding 602 aa. SmEDS1 contained an LP and EP domain. After the resistant eggplant  SmEDS1 gene was silenced   by VIGS, the inoculated eggplant showed wilt at 7 days after inoculation with Ralstonia solanacearum. The result suggested that SmEDS1 positively regulated the eggplant bacterial wilt resistance. The gene expression of MAPK6, RAR1 increased while MAPK3, SIPK, PAD4, SGT1, TGA, EDR1, NPR1, ICS1, GLUA, EIL and HSP90 decreased  after SmEDS1 was silenced. The expression of EBF2 and ACO5  were  not  regulated  by  SmEDS1.  The  results  suggest that EDSI plays an important role in controling eggplant resistance to bacterial wilt.
