| 摘 要： | 目的]通过对5种从不同材料中提取马DNA的方法进行比较研究,寻找最适的提取方法。方法]采用酚仿抽提法、蛋白酶K直接消化法、酸碱裂解法分别从血液、毛发材料中提取马DNA,并通过PCR扩增对提取产物进行检测。结果]通过蛋白酶K直接消化法、酸碱裂解法可从微量血液和毛发中提取到马DNA,均可达到采用酚仿抽提法从血液中提取马DNA的效果,并能满足后续分子生物学试验的要求。结论]通过比较分析,该研究中以酸碱裂解法最佳。

| Abstract: | Objective] To compare five kinds of horse DNA extraction methods from different materials,so as to find the best one.Method] phenol and chloroform method,proteinase K direct digestion method and acid-alkali lysis method were performed to extract DNA from the horse blood and hair respectively,and PCR amplification was used to detect the extracted product.Result] Proteinase K direct digestion method and acid-alkali lysis method can be used to extract DNA from the horse trace blood and hair and able to meet the requirements of the subsequent molecular biological analysis.Conclusion] Through comparative analysis,the acid-alkali lysis method was the best. |
