摘    要:目的]探讨甘蔗试管苗瓶外生根的条件,以期为其瓶外生根技术的开发和应用提供理论依据。方法]以处于增殖培养阶段的甘蔗(新台糖22号)无根试管苗为试验材料,研究了3种不同的外源激素及不同浓度激素处理对该试管苗瓶外生根的影响。结果]一定浓度的赤霉素处理,对新台糖22号无根试管苗基部节的生长有明显的促进作用,并且可以显著提高其移栽存活率,可达到对照组的2.2倍以上,其中以1.5 mg/L赤霉素处理组试管苗的存活率最高,达到83.33%。移栽前用生根粉或萘乙酸浸泡新台糖22号试管苗基部,对试管苗的移栽成活率及其以后的植株生长没有显著影响。结论]该研究结果为甘蔗试管苗瓶外生根技术的应用提供了科学依据。

Effect of Exogenous Phytohormone on ex vitro Rooting of Micropropagated Sugarcane Shoots

Abstract:Objective] To explore the ex vitro rooting of sugarcane shoots,so as to provide theoretical basis for the development and application of its ex vitro rooting technology.Method] Micropropagated shoots of variety ROC22 were cultured on MS medium and the effect of three common exogenous phytohormone on ex vitro rooting was investigated.Result] The culture medium supplemented with GA3 greatly promoted the growth and survival rate of micropropagated shoots.Compared with control,the survival rate treated with GA3 was higher by 2.2 times,especially when treated with 1.5 mg/L GA3,the survival rate reached 83.33%.After cultured on MS medium supplemented with GA3,height and survival rate differed at different concentrations of NAA and ABT,however,the differences were insignificant,which indicated that supplementation of auxins like NAA and ABT before transplanting didn’t have significant effect on survival rate and later growth of sugarcane micropropagated shoots.Conclusion] The study provide scientific basis for the ex vitro rooting for sugarcane micropropagated shoots.
