美女樱叶片离体培养研究

摘    要:以美女樱品种"Quartz"离体叶片为外植体,探讨了不同细胞分裂素、暗培养、硝酸银质量浓度以及不同植物生长调节剂组合对不定芽再生的影响。结果表明:6-BA的诱导效果好于TDZ。在培养基MS+6-BA4.0mg/L+IBA0.15mg/L+蔗糖30g/L上可以使美女樱叶片不定芽的再生率达26.8%。2mg/LAgNO3可以将美女樱叶片的不定芽再生率提高到31.3%,并缩短不定芽发生时间。暗培养5d可以促进美女樱叶片不定芽的再生,活性炭对美女樱叶片离体再生有一定抑制作用。

Abstract:The ideal regeneration system of Verbena hybrida Quartz by leaf explants was established. In this study, the effects of plant growth regulators, dark periods and AgNO3 concentrations on shoot regeneration from leaves of verb ena were investigated. The results showed that the regeneration rate of 268%, was obtained on the medium MS supplemented with 4.0 mg/L 6-BA and 015 mg/L IBA and 30 g/L sucrose. 6-BA was more effective than TDZ in inducing shoot regeneration from leaves of Verbena. 2 mg/L AgNO3 could increase the shoot regeneration rate to 313% and shorten regeneration time.5 days of dark treatment could promote shoot regeneration while charcoal inhibited  shoot regeneration.
