摘    要:为了了解白灵菇对铜胁迫的耐受和应答,为进一步研究铜对白灵菇的毒害机理及避免白灵菇铜污染提供参考。通过综合PDA液体培养试验研究了不同浓度铜(Cu2+,0、50、100、200、300μmol/L)对白灵菇菌丝可溶性糖(蒽酮比色法)、脯氨酸(比色法)含量变化和过氧化物酶(愈创木酚法)、过氧化氢酶(紫外分光光度法)活性变化的影响。结果表明:随着基质Cu2+浓度增加,菌丝体内的可溶性总糖和脯氨酸含量显著降低,当Cu2+浓度达到300μmol/L时,其含量分别下降至对照组的20.94%和35.53%;过氧化物酶和过氧化氢酶活性先是呈升高趋势,后呈降低趋势,活性最大时的铜离子浓度分别是200和100μmol/L。白灵菇菌丝在铜胁迫下可溶性糖和脯氨酸含量没有像高等植物那样随铜浓度升高而先增加后下降的现象;过氧化物酶和过氧化氢酶活性的变化趋势与植物相同,但活性最高的铜浓度比植物偏低,二者说明白灵菇对铜胁迫敏感。在栽培时应注意环境及基质的铜污染程度不能高于100μmol/L。

Influence of Copper Stress on Antioxidant Systems in Pleurotus nebrodensis

Abstract:The objective of this study was to understand tolerance and respond of Pleurotus nebrodensis to copper stress, and to offer a consultation for investigation to mechanisms of copper detoxification and for prevention of copper pollution. P. nebrodensis was incubated in submerged culture medium PDA, which containing different Cu^2+: 0, 50, 100, 200, 300 μmol/L. In hyphae of P. nebrodensis, the content changes of soluble sugar and proline were analyzed by anthrone colorimetry and by colorimetry, respectively. And the activity changes of peroxidase(POD) and catalase(CAT) were assayed by guaiaco method and by ultraviolet spectroscopy, respectively. The results showed that the content of soluble sugar and proline decreased sharply in hyphae with the concentration of Cu^2+increase in medium, and their content were 20.94% and 35.53% lower than the content of the contrast at Cu^2+300 μmol/L, respectively. However, the activity changes of POD and CAT were first increased and then had a decreasing trend with the concentration of Cu^2+increase, their maximum activities were at Cu^2+200 and 100 μmol/L, respectively. In response to copper stress, the activity changes of POD and CAT in hyphae of P. nebrodensis were very similar to that in advanced plant, only Cu^2+ concentration were lower than that in advanced plant at their maximum activities. But the content changes of soluble sugar and proline were different from that of advanced plant, in which content of soluble sugar and proline showed a first increasing and then decreasing trend with the concentration of Cu^2+increase. Therefore,P. nebrodensis was sensitive to copper stress. And a suggestion is put forward that the degree of Cu^2+contamination was no higher than 100 μmol/L for P. nebrodensis cultivation in environment.
