栗属植物cpSSR标记技术的建立与体系优化

摘    要:叶绿体微卫星标记是一种新的分子标记技术。用1对来源于拟南芥的cpSSR引物研究栗属资源,对栗属植物cpSSR反应体系的主要影响因子进行了优化筛选。结果表明,20μL反应体系各组分的最适浓度分别为:2.5mmol/LMg2+,0.5U Taq DNA聚合酶,0.25mmol/L dNTP,10mg/L的DNA模板,正向引物和反向引物都为0.1μmol/L,NTCP40最适退火温度为56℃。该体系已成功应用于栗属资源的遗传多样性研究中。

Abstract:Chloroplast microsatellite is a new molecular marker technology.Experiment was conducted with cpSSR primers obtained from Arabidopsis thaliana for studying the germlasm of genus Castanea Mill.The optimization of main influential factors of cpSSR reaction system in Castanea was studied.The results showed that the optimum concentrations of Mg2+,Taq DNA polymerase,dNTPs,template DNA,and primers in 20 μL reaction system were 2.5 mmol/L,0.5 U,0.25 mmol/L,10 mg/L and 0.1 μmol/L,respectively.56 ℃ was the most suitable annealing temperature of NTCP40 primer.Moreover,it was successful in studying genetic diversity of germplasm of Castanea Mill.
