猪葡萄球菌437-2株的分离鉴定及生物学特性

摘    要:从发生渗出性皮炎的仔猪体内分离到1株细菌,根据其形态特征和生化特性,结合16S rRNA核苷酸序列分析,确定为猪葡萄球菌,并命名为437-2株。用1×109CFU/mL剂量的该菌株经皮下感染小鼠、仔猪,均能引起注射部位皮肤脱毛、破溃、有炎性渗出物、结痂等病变,病理学观察可见表皮局部上皮细胞消失、皮下小血管充血和炎症细胞浸润。应用PCR方法检测分离株携带的表皮脱落毒素相关基因并测序,结果显示该菌株携带表皮脱落毒素ExhD和ShetA基因。药敏试验结果显示该菌株对多种抗生素呈现不同程度的耐药,但对痢菌净、氟苯尼考、头孢曲松钠、阿米卡星敏感。上述结果为猪葡萄球菌引起的仔猪渗出性皮炎的病原学、发病机理及诊断靶标筛选等方面的研究提供了参考。

Abstract:In this study,a strain of bacteria was isolated from piglets with exudative dermatitis.According to its morphological and biochemical characteristics,combined with 16 S rRNA nucleotide sequence analysis,it was identified as Staphylococcus hyicus and named 437-2 strain.Infecting mice and piglets subcutaneously with 0.5 mL(1×109CFU/mL)of this strain could cause skin hair loss,ulceration,inflammatory exudate,scab and other skin lesions at the injection site.Moreover,pathological observation showed that epithelial cells in the epidermis disappeared,subcutaneous small blood vessels were congested and inflammatory cells infiltrated the lesion area.PCR and gene sequencing were used to analyze the epidermal abscission toxin-related genes,the results showed that 437-2 strain carried genes coding ExhD and ShetA,and could express epidermal exfoliating toxin.The results of drug susceptibility tests showed that 437-2 strain exhibited different degrees of resistance to multiple antibiotics.However,it was sensitive to dysentery,florfenicol,ceftriaxone sodium,amikacin.The above results will provide reference for the research of the etiology,pathogenesis and screening of diagnostic targets of piglet exudative dermatitis caused by Staphylococcus hyicus.
