西藏那曲牦牛源产气荚膜梭菌的分离鉴定及生物学特性

摘    要:利用细菌培养、生化鉴定、小鼠致病性试验、PCR技术等方法对西藏那曲市多起牦牛猝死病例进行诊断和分析。结果表明,从病死牦牛脏器中分离得到3株产气荚膜梭菌,命名为AD-01、AD-02和BG-01。其中AD-01和AD-02分离株属于A型,BG-01分离株属于C型。通过对分离株16S rRNA进行序列比对并制作进化树分析发现,AD-01和AD-02分离株为A型产气荚膜梭菌,与中国近年来报道的分离菌株遗传距离较远,属于完全独立的一个分支;同时药敏试验结果显示AD-01、AD-02分离株与BG-01分离株的药敏试验结果有一定差异,产生此现象的原因需进一步研究。本试验成功分离到3株牦牛源产气荚膜梭菌,为牦牛产气荚膜梭菌病的预防和控制提供了理论依据。

Isolation,identification and biological characterization of Clostridium perfringens isolated from yak in Naqu,Tibet

Abstract:We diagnosed and analyzed many cases of sudden death of yak in Naqu district,using bacterial culture,biochemical identification,mouse pathogenicity tests and PCR methods.The results showed that three strains of Clostridium perfringens were isolated in dead or sick yaks,named AD-01,AD-02,BG-01,respectively.The AD-01 strain and AD-02 strain were identified as type A.C.perfringens,while BG-01 was identified as type C.Furthermore,phylogenetic analysis of 16 S rRNA showed that AD-01,AD-02 strain clustered in an independent branch from China isolated strains.Antibiotic susceptibility assay indicated diversity between the AD-01,AD-02 and BG-01,the reason needed to be further investigated.Three strains of Clostridium perfringens from yaks were successfully isolated in this experiment,which provide a theoretical basis for the prevention and control of Clostridium perfringens in yaks.
