黄瓜子叶离体再生体系研究

摘    要:以20个黄瓜自交系2~3天的子叶为外植体,比较了不同植物生长调节物质组合、AgNO3质量浓度和基因型对芽的诱导、增殖、伸长、生根的影响,从而建立了快速高效的黄瓜子叶离体再生体系.结果表明,最佳的培养基配方:诱导培养基为MS 6-BA 1.0 mg/L AgNO3 1.0 mg/L,增殖培养基为MS 6-BA 2.0 mg/L AgNO3 1.0 mg/L,伸长培养基为MS 6-BA 0.1~0.2 mg/L;生根培养基为MS.20种基因型均能诱导出再生芽,诱导率较高的自交系排序:IL 69>IL 103>IL 67>IL 99>IL 77>IL 57,其诱导率依次为94%,88%,87%,78%,70%,62%.

Abstract:The cotyledons from two to three-day seedling have been chosen as explants to establish a high frequent regeneration system in twenty self-bred lines of Cucumis sativus L.. The results showed that the optimum regenerating medium was MS 6-BA 1.0 mg/L AgNO3 1.0 mg/L, the proliferating medium was MS 6-BA 2.0 mg/L AgNO3 1.0 mg/L, the elongating medium was MS 6-BA 0.1-0.2 mg/L, the rooting medium was MS. Twenty genotypes could exhibit shoot regeneration and six genotypes have higher regeneration rate. Their order were IL 69,IL 103,IL 67,IL 99,IL 77,IL 57 from high to low in regeneration rate and the rate was 94%,88%,87%,78%,70%,62% respectively.
