山羊FGF5基因单核苷酸多态性群体遗传学分析

摘    要:为了研究成纤维细胞生长因子5(FGF5)基因在山羊被毛生长中的的作用,进一步寻找与山羊被毛生长相关的遗传标记,为山羊绒毛性状的标记辅助选择和育种提供一定的理论依据,根据人和小鼠成纤维细胞生长因子5(FGF5)基因的同源序列设计引物对山羊基因组进行PCR扩增,将扩增片段进行克隆和测序,并与人和小鼠的成纤维细胞生长因子5基因序列进行同源性比较,确定扩增的片断为山羊的FGF5基因片断。采用PCR-SSCP技术分析了FGF5基因外显子在内蒙古绒山羊、辽宁绒山羊和文登奶山羊品种的多态性,结果表明:FGF5基因在P1和P2引物扩增片段中存在PCR-SSCP多态性,而在P3和P4中没有发现多态。经克隆测序分析,位于外显子1的引物1内存在A→G突变;引物2中发生了碱基序列C→T的突变,这2个突变位点均没有引起编码氨基酸的改变,属于同义突变。对不同山羊品种的基因型和基因频率统计结果表明,引物1中内蒙古绒山羊、辽宁绒山羊、文登奶山羊均以等位基因B为主,且不同群体均处于Hardy-Weinberg平衡;引物2中内蒙古绒山羊、辽宁绒山羊均以等位基因E为主,而文登奶山羊的基因型频率与其他品种有显著差异;内蒙古绒山羊、辽宁绒山羊在引物2位点的基因频率处于Hardy-Weinberg不平衡状态,而文登奶山羊的基因频率处于Hardy-Weinberg平衡状态。

Abstract:In order to study the fuction of FGF5 on goat hair growth and to find the genetic marker of goat hair,it was designed and aimed to identify the single nucleotide polymorphisms(SNPs) of FGF5 gene in various goat breeds including Inner Mongolia cashmere goat,Liaoning cashmere goat and Wendeng diary goat.The 4 pairs of primers for FGF5 gene were designed based on the human and mouse genomic sequence.Two SNPs were identified in the goat FGF5 gene by PCR-SSCP and sequencing.They were in the exon 1 region in different goat lines.In primer 3 and primer 4 no SNP.The sequence data showed that two SNPs found in the exon1 region of the gene gene were A to T and C to T.Population genetic analysis indicated that genotype frequency of the P1 SNPs was similar in different breed,gene B is mean gene.All the population were in Hardy-Weinberg equilibrium state.For primer 2,frequency of the EE genotype in both cashmere goat was higer than in the Wendeng lines,Frequency of allele D in primer 2 in Wendeng goat was higher than that of other breeds.Both cashmere population were in Hardy-Weinberg equilibrium state but Wendeng goat was not in Hardy-Weinberg equilibrium state.
