摘    要:从碱茅(Puccinellia tenuiflora)cDNA文库中分离得到Put-R40g3基因(登录号:AB465547).克隆的cDNA全长为588 bp,其编码的蛋白质共195个氨基酸,与水稻,小麦和拟南芥的R40g3蛋白的氨基酸序列有较高的同源性.对经逆境处理的碱茅幼苗根、叶中该基因的表达分析结果表明,碱茅中Put-R40g3基因的表达有被ABA、盐和干旱诱导的趋势.Put-R40g3蛋白与绿色荧光蛋白(GFP)融合在酵母细胞表达的蛋白质定位研究结果显示了Put-R40g3蛋白存在于细胞质中.同时Put-R40g3基因在酵母中表达,酵母在盐、干旱及氧化逆境条件下提高了对逆境的适应能力.这些结果暗示了Put-R40g3基因是一个与逆境相关的基因.

Abstract:Put-R40g3 gene (accession number: AB465547) was isolated from Puccinellia tenuiflora. The ORF of Put-R40g3 gene is 588 bp which encoded 195 amino acids and showed high similarity to R40g3 protein in Triticum aestivum, Oryza sativa and Arabidopsis thaliana. RT-PCR analyses showed that the expression of Put- R40g3 gene in root and leaf of P. tenuiflora was induced by several abiotic stresses from salts, drought and ABA treatment.
