山羊松果体细胞原代培养及其生物学特性研究

摘    要:目的:观察体外培养山羊松果体细胞的增殖情况和功能状态,为进一步提高季节性繁殖动物的繁殖率提供理论基础。方法:对松果体细胞进行培养,倒置显微镜下观察细胞形态,MTT法检测细胞增殖情况,5-HT免疫组织化学染色显示细胞的功能状态。结果:松果体细胞呈圆形或多角形,折光性强,细胞体积有增大趋势;细胞有明显的增殖现象,大约于11d达到高峰;5-HT阳性细胞数量随细胞的增殖而增加。结论:山羊松果体细胞在体外增殖旺盛。

Abstract:Objective:To observe and understand the proliferation and functional status of goat pinealocyte in vitro. And produce rational for raising reproduction rate of seasonal breeding animal. Methods:Cells were cultured in routine method. Inverted microscope was used to observe the morphologic feature of pinealocyte. Proliferation and functional status of the cells were measured by MTT and 5-HT immunohis-tochemical method. Results:The pineal cells were round, polygon and glazing.And cells would become much bigger with the time going on.The number of the pinealocytes was greatly increased and reached the peak at the 11th day,which was detected by MTT method.Most pinealocytes contained 5 -HT,including many new born ones.Conclusion:Goat pinealocyte could grow vigorously in vitro culture.
