链霉菌SR-11抑菌活性物质的理化性质初步研究

摘    要:从四川各县姜土土壤中分离得到1株放线菌SR-11菌株,经初步鉴定为链霉菌属(Streptomyces).该菌株对姜瘟青枯假单孢杆菌及11种供试菌(其中包括3种病原细菌和8种病原真菌)都有较强抑制作用.对SR-11菌株发酵液的粗分条件和拮抗物粗品的性质进行了研究,结果表明,以体积比为1∶1的乙酸乙酯萃取活性物质最为可行,所得的橙黄色活性粗提物,在37~100℃,pH值3~11处理10~30 min以及胰蛋白酶和蛋白酶K处理1 h后,对青枯假单孢杆菌的抑菌活性不变,说明该物质对热、酸、碱及酶的稳定性强.粗品在官能团反应中,茚三酮反应呈阳性,坂口反应呈阴性,与新霉素的反应结果一致而与链霉素的反应结果相反,说明该物质不是链霉素而可能是新霉素.

Abstract:The strain of SR-11 was isolated from the soil collected from some counties of Sichuan province.By identification,the strain was regarded as Streptomyces.The strain showed antagonism against pathogenic fungus and bacteria.The antibiotic components obtained from Streptomyces SR-11's fermented broth were isolated,and then tested on the physical and chemical stability. The results showed that the antibiotic components of Streptomyces SR-11 were acid-and-alkali-base stable under pH 3~11 and heat-stable treated by 37~100 ℃ for 10~30 min,and maintained the antibiotic activity after treatment of proteinase K and trypsin.Moreover,it was positive in the functional reaction of ninhydrin reaction and negative of Sakaguchi's reaction,which was coincided with those results of neomycin,but contrary to streptomycin.So the antibiotic components could not be streptomycin,in stead,it may be neomycin or a compound.
