杉木优良无性系组培快繁与容器苗培育技术研究

摘    要:以20年生杉木优良无性系根部萌条获得的无菌组培苗作为试验材料,在前人研究基础上,进一步对杉木组培体系进行优化,并开展了指数施肥对组培苗轻基质容器苗生长影响研究,结果表明:最适培养基为DCR+6-BA0.4 mg·L-1+NAA0.02 mg.L-1,其增殖倍数达到4.19,每瓶平均总芽数高达31个,去除顶芽接种方式较之带顶芽,其增殖倍数、有效芽所占百分比分别提高了21.7% ~ 45.5%和31.7% ~ 39.2%;最佳生根培养基为1/4DCR+NAA0.8 mg.L-1,生根率达76.4%,平均生根条数为6.67;不同生根方式对移栽成活率皆无明显差异,均达到90%以上;指数施肥对苗高和高径比影响差异显著,N150为最优.

Abstract:Twenty root suckers were collected as explants to obtain tissue culture plantlets for studying micro-propagation and container seedling propagation of Chinese fir.The optimal media for micro-propagation of shoots comprised DCR + 6-BA 0.4 mg·L-1 + NAA 0.02 mg.L-1,and that for root initiation in micro-propagation comprised 1/4 DCR + NAA 0.8 mg·L-1.The average propagation coefficient was 4.61 and the average number of shoots 3.The method of removing terminal bud inoculation was compared with terminal bud,and the former method resulted in propagation rates and the percentage of shoots number increase by 21.7% ~ 45.5% and 31.7% ~ 39.2% respectively.The rooting rate and the average rooting number was 76.4% and 6.67 with the optimal rooting media.There were no significant differences in transplant survival rates between different ways of rooting,with survivals of up to more than 90% being achieved.The height and height-to-diameter ratio was significantly affected by exponential fertilization,with and the optimal rate being N150.
