摘    要:本实验以牛血清白蛋白为样品进行聚丙烯酰胺凝胶电泳,然后用考马斯亮兰R-250和银染法分步染色。这种复合银染法的灵敏度比单独使用银染法提高约1000倍,比单独使用考马斯亮兰R-250染色法提高约106倍。它在1mm厚的聚丙烯酰胺凝胶上至少可检测出5pg/mm2的蛋白质。在没有放射性防护设施的任何一个生物实验室中,采用复合银染法基本上能够达到或接近放射性标记检测的灵敏度。相比之下,它不仅省去了繁琐的各式各样的标记过程及其放射性污染,同时还具有很高的灵敏度,因此复合银染具有广泛的应用性。

Abstract:Bovine serum albumin was used as sample to carry out polyacrylamide gel electrophoresis and then was stained by using a complex silver stain that included coomassie blue stain and silver stain. The sensitivity of this complex silver stain was 1 000 times higher than silver stain and 106 times higher than coomassie blue stain. At least 5 pg/mm2 protein in gel could be detected. The sensitivity of radioactive label could be reached by the complex silver stain. So the complex silver stain could be a very useful method.\=
