黑芝麻不同外植体离体培养研究

摘    要:以黑芝麻无菌苗的下胚轴、幼根、子叶为外植体,以MS+2,4-D1.5mg/L+ZT0.5mg/L为愈伤组织诱导培养基;MS+6-BA2mg/L-IAA0.5mg/L为分化培养基;1/2MS+0.5mg/LNAA+IAA0.25mg/L为生根培养基进行培养.结果表明,幼根能产生少量愈伤组织,尚未分化出苗;子叶愈伤组织能分化出苗,但分化率低;下胚轴愈伤组织不仅可分化出苗(分化率为36%),而且可形成大量具有分化潜力的胚性愈伤组织,分化绿苗的频率可达67%.

Abstract:Explants were hypocotyls,young roots cotyledons of aseptic seedlings of black seedcoat seame.Induced medium for calli was MS medium with 2,4-D 1.5 mg/L and ZT 0.5 mg/L.Differentiation medium for bud was MS medium with 6-BA 2 mg/L and IAA 0.5 mg/L.Induced medium for root was 1/2 MS medium with NAA 0.5 mg/L and IAA 0.25 mg/L.The results indicated:a few calli formed from young roots in vitro,but there were no buds formed in culture.Some buds were differentiated from calli of cotyledons.Calli from hypocolyls not only differentiated to some buds but also formed a lot of embryonic calli which had differentiation capacity.The differentiation ratio of green shoots could reach to 43%.Hypocolyls were the best explants in the culture of black seedcoat seame in vitro.
