截叶胡枝子组织培养的研究

摘    要:为胡枝子遗传转化奠定基础,对截叶胡枝子的组织培养进行了研究.结果表明以2.1% NaClO对种子灭菌10 min,发芽率较高,污染率为0;初代培养6-BA最佳浓度为1.0~2.0 mg/L,NAA为0.01 mg/L,2,4-D为0~0.01 mg/L,分化系数为2~2.5;继代培养最佳配方为 MS培养基含1倍量的大量元素+6-BA 2.0 mg/L +IBA 0.5 mg/L+2,4-D 0.5 mg/L;生根培养中IBA比NAA更有利于有根苗的生长,随着NAA浓度提高,对生根抑制作用加强,其浓度不宜超过0.1 mg/L,IBA的最佳浓度为0.5~1.0 mg/L.分化苗在培养过程中出现严重的黄化现象,针对这一问题,开展了无机盐(NH4NO3、MgSO4·7H2O、CaCl2·2H2O)、铁盐、糖的浓度和光的种类的比较试验.结果表明,氮、镁、铁对黄化有显著影响,当MgSO4·7H2O浓度为300 mg/L、NH4NO3浓度为1 600 mg/L、MS中铁盐浓度为3倍时,黄化苗率最低,而其他3种因素在该试验不同处理间的差异不显著;6种因素各处理间在子叶节的分化系数上的差异均不显著.

Abstract:In order to develop the genetic transformation, tissue culture of Lespedeza cuneata was studied in this paper. The results indicated that the best time of seed disinfectant of 2.1% NaClO was 10 min, and the pollution percentage was zero under this sterilization condition. The optimal concentrations of primary differentiation were 6-BA 1.0-2.0 mg/L,NAA 0.01 mg/L and 2,4-D 0-0.01 mg/L, on which the differentiation index could reach 2-2.5 The optimal culture medium in secondary differentiation was MS medium containing original macroelements+6-BA 2.0 mg/L+IBA 0.5 mg/L+2,4-D 0.5 mg/L. IBA was more suitable than NAA for the rooting of plantlets. The optimum concentration of IBA was 0.5-1.0 mg/L, whereas that of NAA should be less than 0.1 mg/L, since high concentration of NAA had an inhibiting effect on rooting. In order to solve the serious problem of bud etiolation, experiments with different concentrations of NH4NO3, MgSO4·7H2O, CaCl2·2H2O, iron salt, sugar and types of lights were conducted. The results showed that the concentration changes of N, Mg and Fe had significant effects on inhibiting etiolation, and the optimal concentrations were NH4NO3 1 600 mg/L, MgSO4·7H2O 300 mg/L, and iron salt 83.4 mg/L. Another three factors had no significant effects on etiolation. And there were no significant differences in the generation index for all of the six factors in the etiolation experiments.
