响应面法优化莱氏绿僵菌SL200714菌株液态发酵培养条件

摘    要:莱氏绿僵菌(Metarhizium rileyi)是一种广泛分布并对夜蛾科类害虫具有高致病力的虫生真菌。为优化莱氏绿僵菌液态发酵培养条件,进一步提升发酵液生物量。本研究首先通过单因素试验探究3种不同类型培养液对莱氏绿僵菌SL200714发酵液生物量的影响,利用Plackett-Burman对主要影响生物产量因子进行筛选,通过最陡爬坡试验确定关键因子适宜范围,最后通过响应面法建立莱氏绿僵菌SL200714液体发酵菌丝生物量多元回归方程。结果表明,莱氏绿僵菌SL200714菌株利用SDY培养液在培养温度为25.08℃,摇床转速为182.11 r/min,装液量为100.03/250 mL条件下进行液态发酵为理论最佳培养条件。在此条件下,每10 m L发酵液可产菌丝生物量0.361 g。本研究利用响应面法对莱氏绿僵菌液态发酵培养条件进行优化,提高莱氏绿僵菌发酵液每毫升生物量,为后续工厂化生产莱氏绿僵菌分生孢子粉奠定基础。

Abstract:Metarhizium rileyi is a widely distributed entomopathogenic fungi with high pathogenicity to noctuid pests. The present study aims to optimize the liquid fermentation conditions of Metarhizium rileyi, and further increase fermentation broth biomass. In this experiment, the single factor experiment was selected for evaluating the effect of three culture solutions on fermentation broth biomass of Metarhizium rileyi, and screened the main factors affecting bioyield by Plackett-Burman. The suitable range of key factors was determined by the steepest climbing test. The multiple regression equation of mycelial biomass in liquid fermentation of Metarhizium rileyi SL200714 was established by Response Surface Method. The results showed that the strain of Metarhizium rileyi SL200714 was subjected to liquid fermentation using SDY medium under the conditions of culture temperature of 25.08℃, rotating speed of 182.11 r/min, and liquid volume of 100.03/250 mL. In this optimal culture solution, 0.361 g mycelial biomass could be produced per 10 mL fermentation broth. Response surface methodology was used to optimize the liquid fermentation culture conditions of Metarhizium rileyi, and to improve the biological yield per milliliter of Metarhizium rileyi fermentation broth, and the study could lay a foundation for the subsequent industrial production of Metarhizium rileyi conidial powder.
