紫外线、He-Ne激光对葡萄白腐病菌拮抗菌PT2的复合诱变效应

摘    要:采用紫外线、He-Ne激光及两者复合诱变的方法,分别处理对葡萄白腐病有拮抗作用的枯草芽胞杆菌PT2,以提高其抑菌效果。结果表明,紫外线和He-Ne激光单独作用,所筛菌株UV10与HN14抑菌效果提高率分别为15.0%与21.7%。而经过对复合诱变条件确定,在紫外线照射90 s后,再用15 mW的He-Ne激光辐照10 min,筛选出最佳正变菌株F15,抑菌效果提高30.9%,明显高于单因素诱变,经25代传代试验证明其拮抗能力可以进行稳定遗传。

Abstract:The strain PT2 Bacillus subtilis, which has an antagonistic ability to Coniella diplodiella Petrak &; Syd. was irradiated respectively by UV and He-Ne laser, and the mutant strains UV10 and HN14 were obtained. Both of them increased the antagonistic ability by 15.0% and 21.7% compared with PT2. The results indicated that the coordinative effect of UV and He-Ne laser was better than that by using them individually. The cells were treated firstly by UV for 90 seconds and then by He-Ne laser for 10 minutes in 15mW. The best positive mutation strain F15 was obtained, whose antagonistic ability was 30.9% higher than PT2’s. By cultivating it from 1st to 25th generation, its antagonistic ability proved to be stable in heredity.
