基于ITS序列海带目种质资源的物种鉴定研究

摘    要:为了更准确地鉴定海带目种质资源,为种质保存和保护提供依据,研究采用海带目的核糖体DNA内转录间隔区(Internal Transcribed Spacer,rDNA ITS区)的通用引物,以待鉴定海带目样品的基因组DNA为模板通过PCR方法扩增出ITS序列,在测序的基础上对25棵分别采自日本和烟台海区的海带目样品进行分子鉴定。结果表明,25个待鉴定褐藻目样品和16个已知物种,通过ITS序列聚类分析,分别聚在不同的单系类群中且在物种鉴定树中种内的支持率均较高,分属于2科3属,聚类为7个不同的种,形态学观察的结果与分子鉴定结论相吻合。海带目ITS序列引物通用性强,有效扩增率高,可用于海带目物种的快速鉴定。

Identification of Laminariales Germplasm Resources Based on Ribosomal DNA Internal Transcribed Spacer (ITS) sequence

Abstract:The aims were to identify Laminariales germplasm resources accurately and provide the basis for germplasm conservation and protection. The universal primers of ribosomal DNA Internal Transcribed Spacer(Internal Transcribed Spacer of rDNA, ITS) were adopted to amplify the ITS sequences of the undetermined samples by PCR, on the basis of the sequencing, the molecular identification was carried out among 25 samples collected from Japan and Yantai sea area. The results showed that 25 undetermined samples and 16 known species were clustered in different groups by clustering analysis of ITS sequence. Within the species identification trees, the ratings are high. The 25 undetermined samples were clustered in three genera, two families and seven different species respectively. The results of morphological observation conformed to the molecular identification. The primers of Laminariales ITS sequence could be used in rapid species identification as its versatility and high efficient amplification rate.
