岷江百合生境及遗传多样性

摘    要:在踏查岷江百合分布区的基础上随机选取两个阴坡与阳坡群体,设样地调查其群落特征,测定其土壤化学性质指标,并用ISSR分子标记分析两群体遗传多样性。结果表明,岷江百合生长在兰花莸、小角柱花、美丽胡枝子、黄花蒿等为优势的干旱河谷灌丛中,在垂直结构上居于灌木层片。数量上较多,但因冠副较小,群落外貌上不明显。土壤为褐土,呈弱碱性,土壤潜在肥力水平较高,但有效成分较低。阴坡土壤有效成分高于阳坡。物种水平的多态性谱带的比例为96.72%,有效等位基因数为1.5572,Shannon’s多样性指数为0.4932,平均期望杂合度为0.3276。其中阴坡群体的遗传多样性参数要高于阳坡群体。群体间遗传差异占总遗传差异的17.57%,群体内遗传差异占总遗传差异的82.43%。两个群体间的基因流为5.0660,说明两群体亲缘关系较近,而群体内遗传分化强烈,阴坡群体的遗传分化强于阳坡群体。

Abstract:The characters of two populations randomly selected based on a survey in the distribution area of Lilium regale were investigated,and the soil chemistry properties and the genetic diversities of the two populations were also analyzed by means of ISSR molecular marker.Results show that L.regale grows in dry valley shrubbery dominated by Caryopteris incana,Ceratostegma minus,Lespedeza formosa,and Artemisia annua,and locates at shrubbery synusia in vertical structure.Although there are large number of individuals in community,L.regale is unobvious to be seen by community appearance due to its small crown.L.regale grows on alkalescent cinnamon soil with high potential fertility and low available fertility.The available fertility of the soil at shady slope is higher than that at sunny slope.At the species level,the proportion of polymorphic loci is 96.72%,the effective number of allele(Ne),Shannon diversity index(I)and the average expected heterozygosity are 1.557 2,0.493 2 and 0.327 6,respectively.The genetic diversity index of shady slope is higher than that of sunny slope.The result of AMOVA analysis indicates that the variation within population accounts for 82.43 percent and the variation among populations occupies 17.57 percent,and the gene flow is 5.066 0.It demonstrates that the relationship between the two populations is closer.Gene differentiation is acute within population,especially for the populations at shady slope.
