摘    要:LACS(long-chain acyl-CoA synthetase)编码长链酰基CoA合成酶,能把游离脂肪酸转化成长链酰基辅酶A,在油脂代谢过程中有重要的作用。对向日葵HaLACS7编码蛋白进行生物信息学分析,预测HaLACS7编码蛋白为酸性蛋白且为亲水性蛋白,蛋白二级结构为无规则卷曲,亚细胞定位于过氧化物酶体。经系统进化分析发现,HaLACS7与莴苣(Lactuca sativa L.)和洋蓟(Cynara cardunculus var. scolymus L.)的LACS6进化关系最近,序列相似性均为84%,且与拟南芥LACS6LACS7聚在一个大分支上。HaLACS7在向日葵中组织的表达量大小为舌状花>根>种子>管状花>叶>茎。HaLACS7在高、低油酸向日葵种子发育后期表达量均较高,但2份材料种子在不同发育时期表达变化趋势明显不同,推测HaLACS7的表达可能影响向日葵种子油酸的合成。

Abstract:LACS encodes for long-chain acyl-CoA synthetase, a protein that converts free fatty acids into long-chain acyl-CoA, an essential lipid metabolite. The bioinformatics analysis of the HaLACS7 encoded protein in sunflower was carried out in this study. The protein encoded by HaLACS7 was expected to be an acidic and hydrophilic protein. The protein encoded by HaLACS7 had an irregular coil secondary structure, and it’s subcellular location was anticipated to be in the peroxisome. With 84% sequence similarity, HaLACS7 was shown to be closely related to Lactuca sativa L. and Cynara cardunculus var. Scolymus L. LACS6, and clustered in a broad branch with LACS6 and LACS7 of Arabidopsis thaliana L. The expression level of HaLACS7 in sunflower tissues was ligulate flowers > root > seed > tubular flower > leaf > stem. The expression level of HaLACS7 was higher at the late development stage of high and low oleic acid sunflower seeds, but the expression trend was significantly different at different seed development stages of the two materials. Therefore, it was suggested that the expression level of HaLACS7 may affect the synthesis of oleic acid in sunflower seeds.
