茉莉酸甲酯对罗汉果SQS、CS和CAS基因表达的影响

摘    要:以人工授粉后50d的罗汉果果实为试材,喷施不同浓度的茉莉酸甲酯(Methyl Jasmonate,MeJA),诱导不同时间后取样并提取RNA,采用实时荧光定量PCR测定罗汉果甜苷V代谢途径3个关键酶(角鲨烯合酶:squalene synthase,SQS;葫芦二烯醇合酶:cucurbitadienol synthase,CS;环阿乔醇合酶:cycloartenol synthase,CAS)基因的相对表达量,以期为掌握SQS、CS和CAS的表达规律,进而为提高罗汉果甜苷V的代谢提供参考依据。结果表明:MeJA对SQS和CS的表达均有促进作用,在200μmol·L-1的MeJA诱导后12h和24h,SQS和CS的相对表达量最高,分别为处理前的11.12倍和9.82倍;MeJA能抑制CAS的表达,在100μmol·L-1的MeJA处理后6h,CAS相对表达量降至最低,仅为对照的0.13倍。

Abstract:The fruits of Siraitiagrosvenoriiafter fertilization for 50 days were used as materials,treated with different concentrations of methyl jasmonate(MeJA).After inducing with MeJA for different times,RNA was extracted from the sample fruits.Expression levels of squalene synthase(SQS),cucurbitadienol synthase(CS)and cycloartenol synthase(CAS)(the genes encoding the key enzymes of mogroside V biosynthetic pathway in S.grosvenorii)were detected by quantitative realtime PCR in order to provide theoretical basis for improving mogroside V metabolism by grasping the rule of SQS,CSand CASexpression.The results showed that the expressions of SQSand CS were increased by the induction of MeJA;12hours and 24 hours after treatment with 200μmol·L^-1 MeJA,the expression of SQSand CS were the highest,accounting for 11.12 folds and 9.82 folds of that before treatment,respectively.However,the expression of CAS was decreased by the induction of MeJA;6hours after treatment with 100μmol·L^-1 MeJA,the expression of CAS was the lowest,accounting for only 0.13 folds of that before treatment.
