黑果小檗中总黄酮的超声提取及其含量测定

摘    要:目的]建立一种测定黑果小檗中总黄酮含量的方法。方法]用超声波提取黑果小檗果实中的总黄酮,然后以芦丁为标准品,采用分光光度法测定总黄酮含量。结果]芦丁浓度与吸光度的线性关系方程为A=0.010 74 C+0.009 214(r=0.999 2),表明芦丁在7.5~60.0μg/ml范围内与吸光度值线性关系良好,平均回收率为93.6%;测得黑果小檗果肉、籽粒中总黄酮的含量分别为0.78%和1.89%。结论]该方法快速、简便、可靠,可用于黑果小檗中总黄酮含量的测定。

Extraction of Total Flavonoids in Breberis heteropoda Shrenk by Ultrasonic and Determination of Its Content

Abstract:Objective] The aim was to establish a method to determine total flavonoids content in Breberis heteropoda Shrenk.Method] Ultrasonic technology was used to extract the total flavonoid of the fruits of B.heteropoda Shrenk,and then with rutin as the standard substance,total flavonoids were determined by spectrophotometry.Result] The results showed that the linear relationship equation of rutin concentration and the absorbance was A=0.010 74 C+0.009 214(r=0.999 2),which indicated that there was a good linear relationship between the absorbance and the concentration of rutin within the range of 7.5~60.0 μg/ml.The average recovery rate was 93.6%,and the contents of total flavonoids in pulp and seed were 0.78% and 1.89% respectively.Conclusion] The method was found to be fast,simple,reliable and good for the determination of total flavonoids in B.heteropoda Shrenk.
