脾脏转移因子提取影响因素的研究

摘    要:研究了改变转移因子冻融提取法中的冻融次数、透析时间及匀浆介质等因素对转移因子收率的影响。通过对转移因子的有效成分多肽和核糖核酸的提取浓度的测定,发现用去离子水做匀浆介质所提取的多肽和核糖核酸效果较好,反复冻融6次转移因子收率最好,在一定范围内,随着冻融次数的增加,透析液中的多肽和核糖核酸浓度逐渐增加,透析24h多肽浓度趋于稳定,透析12h核酸浓度达到高峰。

Abstract:Through determining the concentration of polypeptide and RNA,the influence of different freezen times,method of thrawing on the yield of pig spleen transfer factor was discussed in this experiment.Higher concentration of peptide and RNA were detected when spleen tissue was homogenized with deionized water than that with normal saline,indicating that higher diversity if osmotic pressure was more effective in breaking cell membrane to release peptide and RNA from the the treated cell.The highest TF yield coul...
