三-(2,3-二溴丙基)异氰脲酸酯对斑马鱼胚胎发育的毒性研究

摘    要:为了探究三-(2,3-二溴丙基)异氰脲酸酯(TBC)这一新型含溴阻燃剂对斑马鱼胚胎的毒性效应及潜在的致毒机制,将受精后2h内的斑马鱼胚胎分别暴露于0.2、1、5、25μg/mL的TBC和0.1、0.5、2.5、12.5μg/mL的17β-雌二醇(E2)以及各浓度TBC+2.5μg/mL E2中,分别在24、48、72、120hpf对胚胎进行形态学观察并统计。发现TBC对斑马鱼胚胎72hpf孵化率的抑制作用及120hpf致畸作用呈剂量效应关系,对120hpf胚胎的致死作用较小;E2对斑马鱼胚胎的孵化率亦有抑制作用;在120hpf,TBC和E2对斑马鱼胚胎均有致畸效应但致畸形态有差异;TBC的EC50为1.248μg/mL,E2的EC50为0.968μg/mL及LC50为2.834μg/mL。结果表明,TBC在一定程度上可抑制雌激素对斑马鱼胚胎的毒性效应,呈现潜在的抗雌激素效应的内分泌干扰作用。

Abstract:In order to explore the toxic effects and potential toxic mechanism of tris-(2,3-dibromopropyl) isocyanurate(TBC) ,a new brominated flame retardant, on the zebrafish embryos. The zebrafish embryos fertilized within 2 hours were exposed by TBC(0.2,1,5, and 25 μg/mL), E2 (0. 1,0. 5,2.5 and 12. 5 μg/ mL)and various concentrations of TBC+2.5μg/mL E2. The embryos were observed and counted by a low magnification microscope at 24, 48, 72 and 120 hpf. The toxic effects of TBC on zebrafish embryonic development were evaluated by hatchability, LCso (concentration for 50% of lethal)and ECs0 (concentration for 50% of maximal effect). The results showed that TBC caused the inhibition of hatchability at 72 hpf and the teratogenic effects at 120 hpf on zebrafish embryos with a dose-dependent manner and the embryo lethal effect of TBC was less at 120 hpf; E2 inhibited the hatchability of the zebrafish embryos at 72 hpf TBC and E2had teratogenic effects on zebrafish embryos with different teratogenic pattern at 120 hpf; the EC50 of TBC was 1. 248 μg/mL, the ECs0 and LC50 of E2 were 0. 968μg/mL and 2. 834 μg/ml, respectively. It indicated that TBC inhibited the toxic effects of estrogen on zebrafish embryos in a certain degree, and showed potential endocrine disruption function with anti-estrogenic effects.
