摘    要:以铁皮石斛未成熟种子为外植体,在离体条件下进行原球茎诱导,类原球茎增殖、分化、生根试验研究。结果表明,以种子萌发直接诱导原球茎的适宜培养基为MS+1.0mg/L BA+0.5mg/L NAA+0.5g/L活性炭(AC)+1.0g/L水解酪蛋白(CH),诱导率达95%以上。在MS+1.0mg/L BA+0.5mg/L KT+0.5mg/L NAA+0.5g/L AC+0.6g/L CH条件下可获得类原球茎最大增殖系数(达18.7),该培养基也适宜于维持类原球茎的增殖保存。类原球茎的最大分化率为93.7%,所需的培养基成分为MS+0.5mg/L BA+0.5mg/L KT+0.2mg/L NAA+0.5g/L AC+0.6g/L CH。试管苗在1/2 MS+0.5mg/L NAA+0.5g/L AC培养基上生根率达100%,且根系健壮发达,移栽后全部成活,长势良好。

Abstract:To establish and optimize the technological system for in vitro propagation of Dendrobium candidum, the immatured seeds used as explants were applied to conduct rapid in vitro breeding of D. candidum. The results showed that the optimal medium for seeds germination and the protoeorm induction was MSff- 1. 0 mg/L BA+0. 5 mg/L NAA+0. 5 g/L AC (activated carbon)+1. 0 g/L CH (casein hydrolysate),and the induction rate was above 95 % The maximum propagation coefficient of protocorm- like-body (PLB) proliferation was 18. 7, which was obtained on the condition of MS+1.0 mg/L BA+0. 5 mg/L KT+0. 5 mg/L NAA+0. 5g/L AC+0. 6 g/L CH, and this medium was also suitable for the maintenance of PLBs. After transferred to MS+0. 5mg/L BA+0. 5 mg/L KT-F0. 2 mg/L NAA+0. 5 g/L AC-f-0. 6 g/L CH, PLBs developed into adventitious shoots and the highest differentiation rate was 93.7%. For rooting, adventitious shoots were transplanted on 1/2 MS medium. supplemented with 0. 5 mg/L NAA and 0. 5 g/L AC,the rooting rate was up to 100% and roots developed well. All of the plantlets survived and grew well after transplanted into the soil in the greenhouse.
